We are going to do some maintenance on our database on Friday December 13th from 16:00 UTC until 20:00 UTC. During this time new tasks will not be distributed and your client will not be able to report completed results. However, your client will automatically retry later and be able to retrieve new work and report completed results once the maintenance is completed.

The website and forums will be available during this work.

Mapping Cancer Markers Beta Test - Version 7.27
We will be starting a new Beta test for Mapping Cancer Markers to help with checkpoint issues that members have noticed. Involved with this beta test will be additional logging.

Note: We will have assimilation turned off for this beta test which means results may stay in your Result Status page longer than usual.

This beta test will have 50,000 work units which will mean 100,000 results sent out.

Mapping Cancer Markers Beta Test - Version 7.28
We will be starting version 7.28 for beta testing soon. This will include a few fixes, one of which will be addressing the large number of invalids. This test will only have 20,000 work units to start.

Limited Tasks in Progress for Newly Connected Devices

Recently there has been an increase in devices that attach to World Community Grid and pull down the maximum number of allowed tasks (5 * number of processing cores). Some of these devices disappear quickly and never return any of the tasks they there were assigned.

Those assigned tasks time-out and are then assigned as high priority tasks to other users. Due to the restrictions on who can be assigned these high priority tasks and the rate at which they are being created we are having troubles distributing work efficiently.

In order to reduce this issue, we are instituting a new policy where newly connected devices will only be allowed to have a maximum of tasks in progress equal to the number of processing cores the devices has. This special limit will be in place for only 3 hours after the device is connected. Then the normal limits will apply.

As part of this we are currently experiencing issues with effectively distributing work. We have over-scheduled workunits that were going to timeout and set these tasks so that they are not assigned as high priority tasks. However, they are still associated with a platform (Windows, Linux, etc). Volunteers will see an unusually larger number of one of the following two messages over the next several hours while this backlog is reduced:

Message #1: Message from server: We are currently experiencing high load and are temporarily deferring your scheduler request. Your client will automatically try again later.

Message #2: Tasks are committed to other platforms

We apologize for these challenges as we adapt to the new participation patterns of some of our new volunteers. We expect this change to largely mitigate this situation.

Unplanned 2 Hour Outage Today [Resolved]

Due to a misconfiguration error, the website was unable to respond file uploads, scheduler requests or https communications. Users attempting to access the site would have received a message stating that the service was not available.

We have corrected this misconfiguration and the application is now working properly again.

Beta Test for Content Delivery Network
In a few minutes, we are starting a beta test to test a change in how some files that the software downloads, are delivered to you. This will help us continue to handle future growth as well as the occasional spike in download requests that periodically occurs.

We are going to use SoftLayer's Content Delivery Network (http://en.wikipedia.org/wiki/Content_delivery_network) to support the download of certain files that are frequently downloaded. Most users will not notice a change as a result of this change. However, for users whose firewalls are set to allow the World Community Grid – BOINC software to download files from only specific IP addresses, you need to update your firewall configuration as per the instructions below.


Technical details for those needing to make change their firewall configuration
Currently, the files in question are downloaded from the URL: https://download.worldcommunitygrid.org, which is associated with the IP addresses 170.224.194.69 and 170.224.160.205.

After this change, these files will be download from https://BDD7.https.cdn.softlayer.net instead. With a Content Delivery Network, this URL is associated with a list of IP addresses, which is included at the bottom.

Check to see what IP address range you are likely to use at your location by typing the following command into a command prompt window:

Database Change - No work sent or received on Jan 24 from 15:00 UTC - 19:00 UTC
We are going to making a change to the configuration of the database that supports the grid. This will result in improved performance of the database. However, in order to implement the change we have to stop access to the database for up to 4 hours. As a result, you will not be able to report completed jobs or fetch new jobs to process during this work.

We apologize for any inconvenience that this causes.

We have encountered some batches of work for Mapping Cancer Markers that are only generating errors on the volunteer computers. We have temporarily stopped sending work for the project while we investigate the issue.

Webcast Summary
In her live presentation, Professor Carbone explained the overall aims of the project: to better understand intracellular protein interactions and to identify the specific circumstances that cause degenerative neuromuscular diseases. Muscular dystrophy is probably the best-known, but there are hundreds of these diseases, in which muscles progressively weaken because of improper functioning of muscle cells themselves or associated nerve cells. To find better treatments, researchers first need to understand the interactions between thousands of different proteins potentially associated with the disease. World Community Grid members have been absolutely essential to this effort; nearly 300,000 members donated computing time to help her team's efforts.

In Phase 1, the project screened 168 well-understood protein pairs to demonstrate the accuracy and usefulness of computational modeling for identifying pairs of proteins that bind together, which is an essential step to finding better treatments. Proteins are very difficult objects to simulate because they have complex 3D structures and the number of possible orientations is immense. However, the proteins were not modeled at random; the research team used knowledge about the proteins' evolutionary history to predict interaction sites and possible partnerships.

The team found that it was possible to accurately predict binding sites on the proteins, meaning that they could simulate a smaller number of positions for each pair, greatly increasing the project's efficiency without significantly reducing the accuracy of the results.

Because of these advances, Phase 2 of the project could analyze pairings of 2,246 proteins -€“ this raw data was completed in 2013 and the analysis is currently underway. The team hopes to have a full database of information about the interactions between these proteins within 18-24 months.

There are still many steps in the path to an effective treatment or cure for muscular dystrophy, but the detailed and extensive knowledge generated by World Community Grid members will be a tremendous resource to Professor Carbone'€™s team and to others who are working toward these goals.

New hope in the fight against childhood cancer

Blog post announcing a breakthrough in the search for a new treatment for neuroblastoma, written by Dr. Akira Nakagawara, principal investigator for Help Fight Childhood Cancer


Today, thanks to advances in modern medicine, 80% of children diagnosed with cancer are cured. But the prognosis is not nearly as good for children with neuroblastoma, the most common form of cancer diagnosed in infants. Neuroblastoma is a tumor of peripheral nerve tissues that often starts in the adrenal glands and sympathetic ganglia of the neck, chest, or abdomen, and affects approximately one in 8,000 children in the United States and Japan.

More than half of neuroblastoma cases are classified as high risk, and only 30% of these children are cured – a rate that has not improved for two decades. New treatments are urgently needed for this dangerous disease.

Our research team at the Chiba Cancer Center in Japan has been working to develop a new treatment for neuroblastoma. With the help of volunteers participating in the IBM World Community Grid initiative, we have just discovered seven new drug candidates that could potentially be used in new medicines that fight childhood neuroblastoma. These drug candidates work by activating a self-destruct mechanism present in neuroblastoma cancer cells, killing them without affecting healthy cells.

Neuroblastoma cells have a receptor on their surfaces called TrkB. When molecules bind to the TrkB receptor and inhibit its function, a tumor suppressor gene called p53 is activated, causing the neuroblastoma cell to self-destruct in a process called apoptosis. Apoptosis is one of the body's natural processes, and ordinarily helps to eliminate damaged cells before they can form a tumor. However, the TrkB receptor in neuroblastoma suppresses this self-destruct function. A similar TrkB process is involved when many adult cancers, including breast, lung, pancreatic, prostate, and colon cancers, metastasize (i.e. spread beyond an initial site). This means our findings may have implications for treating adult cancers as well.

Our strategy was to search for small molecules that would bind to and inhibit the TrkB receptor on cancer cells. There were millions of potential molecules to examine, making it infeasible to synthesize and test each of them in the laboratory. Instead, we partnered with World Community Grid to create the Help Fight Childhood Cancer project, using computer-based modeling on a massive scale to conduct this search. With the help of over 200,000 volunteers around the world contributing their spare computing power, we screened three million molecules in just two years – a process that would have taken more than 55,000 years on a single computer – and identified seven promising drug candidates for further study.

After additional laboratory testing, we have discovered that the seven drug candidates are very effective at destroying neuroblastoma tumors in mice, even at very low dosages, with no immediately apparent side effects. These results have been published in the peer-reviewed journal Cancer Medicine, available online since January 2014.

Based on these very promising findings, we are now seeking a pharmaceutical partner to collaborate on the further development and testing needed to produce an approved medicine.

This breakthrough was made possible by the support of volunteers around the world who donated their computing power through World Community Grid. On behalf of our research team, I would like to thank World Community Grid volunteers from the bottom of my heart.

借助IBM的WCG平台,我们发现了7种新的潜在药物,可以在不影响正常细胞的前提下,消灭癌细胞。
神经母细胞瘤细胞表面有个TrkB受体,一旦有分子和该受体结合,癌细胞就会自毁。因为大多数癌细胞(例如:乳腺癌、肺癌、胰腺癌、前列腺癌、肠癌以及癌细胞扩散)都和TrkB受体有关,所以我们的药物不仅可以用于神经母细胞瘤治疗,也很有可能用于其他癌症治疗。
在其后的实验室测试中,我们发现这7中候选药物,只用很小的剂量就能杀死实验鼠体内的神经母细胞瘤,而且没有大都副作用。研究结果论文发表在2014年1月的《癌症药物》杂志上了。
据此研究结果,我们现在正在寻找合作制药厂商,进行后续的开发及测试。

World Community Grid will be performing standard maintenance on our website Sunday, 9 March from 2:00 UTC - 8:00 UTC. During this time the website and forums will not be available.

Additionally, volunteers will not be able to attach new devices to World Community Grid. However, existing devices connected to World Community Grid will be able to report completed results and fetch new work without interruption.
大意:
3月9日,10至16点将对网站进行例行维护,届时网站可能无法正常访问。注:除新用户注册外,BOINC服务不会受影响。
